Fresh rebuild, now wont start?

Just recently finished up my 416 rebuild. when the wheeler was at the shop that it was worked on at i was riding it around and it was decent just needed some carb adjustment but the last few times ive messed around with it i havent even been able to get it to try to start. it just sits there and cranks over all day. i moved the needle clip up and down one from stock and both just made it worse. i currently have a 168 main in running without the air box lid. any insight on why i cant even get it to start would be helpful

Determine the following and report back:

1. Its getting fuel, will it start with starting fluid?
2. Change the plug, check for spark at same time
3. Check/adjust the valves to spec.

All of that checks out we got bigger problems on our hands.
