2005 ltz 400 with innards removed from muffler and lid off. Elevation 3500'. I first tried a 142.5 jet and it ran fine. plug was clean, but a little white. Now has a 145 main and still runs good, plug still a little white. 25 pilot. If it ran fine with the 142.5 and i bumped it up to a 145 am I safe as far as not being too lean? also, do i need to do anything with the needle?

you could probley go a little higher on your main you want your plug to be a light brownish color and if its still running ok you might not need to do anthing to your needle

and the fuel adjustment screw...should it be adjusted any? does it control the 1/4 to 3/4 throttle only?

yeah you may need to make a small adjustment to your air/fuel screw but you need to remember where its at now so you can put it back if you need to and only make small adjustments at a time, and yes 1/4 to 3/4 only
