It's time for begining of the seaason maintainence! I change the plug and the motor oil and clean the air filter on my 2005 250ex twice a year but i have never changed the grease in the rear end or differential or whatever you call it.

Is this difficult to do? Do i need to do it? What type grease does it take?

Also, the motor oil... I use 10w-40 atv oil from a local kawasaki dealer where i live. Is this ok to use? Could i buy a synthetic like royal purple and put in there or would that be a bad idea? What do you use?

there should just be a grease fitting and u put a grease gun on it and pump grease into it untill u see it coming out in places then stop and u should be ok.
and the oil, it really doesnt matter what kind u use but make sure its the right weight 10-40 sounds right and u can use a synthetic i have in the past and had mixed results but i use honda oil they sell regular and synthetic i think the honda oil is the best but thats just me syntetics will last longer and usually stay cleaner.
