will a KEIHIN Carburetor . PWK72RAA GD 40mm be an improvement over a stock carb on an EX?

A little. Dont change the carb. A 400ex has a big enough carb stock. Mod the motor first a carb should be the last thing on your list. if you have it port and polished/cam/exhaust/valves/k&n/high compression/ then add a big carb.
