what would be the best upgrade to get on a 400ex that would increase power

NOS!!!!!:D no im just joking unless you want to get it, but the best thing to get would be a full system exhaust and header, but to me thats the best thing to get to hop up on power.

I don't recommend you NOS if you don't drag.The NOS is hard on all part of the motor (head gasket,piston,connecting rod,crank,clutch,tranny ... )

Get a power-up kit. Moose works and K&n are common producers. The Moose kit contains a K&N airfilter (and prefilter), and a rocking jet kit for the carb. While your at it, have a whitebrothers E-Series pipe installed. The E pipe will provide more low-end accleration (as will the FMF). But a Yoshimura or Fat Boy will provide more top end speed.

BE SURE TO RE-JET.

You will not believe the difference. I mean wow. You'll have to learn to ride all over again.
