wierd, i've ridden both

handling - Z400
speed - 250R hands down, bottom end goes to the 4 stroke though.
jumping - hrmmm tie, both stock suspensions suck for landings. both are good jumpers and are easy to get air on. though the Z400 tends to jump nose high more often than the 250R.

both are highly modifiable with lots of parts out there. safe side says go for the 4 stroke, while wild side says get the nut pulling speed of a modified 2 stroke...if yer thinking about modding them.

Z400 are great bikes but can't give you the thill of a two stroke powerband.
